Rapid Visual Detection of Pathogenic Streptococcus suis Type 2 through a Recombinase Polymerase Amplification Assay Coupled with Lateral Flow Test

Objective: Streptococcus suis serotype 2 (SS2) is an important zoonotic pathogen causing serious disease and even death in pigs humans. Public health events economic losses caused by SS2 have prompted widespread concern. Because of the unavailability vaccines, development rapid detection methods for timely diagnosis infection or contaminated products, monitoring its prevalence susceptible animals populations, required to aid prevention control infections. Methods: Several sets primers one probe a recombinase polymerase amplification (RPA) assay targeting cpsJ2 gene were designed synthesized. Lateral flow (LF) tests combination with RPA used provide visual results. Primers high efficiency screened, reaction system was optimized. Indicators effectiveness evaluated. Results: The established method had limit 100 copies/reaction recognizing rather than other organisms. sensitivity 100%, as evaluated infected animal samples. could be completed within 20 min only constant temperature equipment. Conclusion: rapid, visual, sensitive specific RPA-LF showed superior performance expected widely applied fight resource-limited areas.
